Abstract
Endo-xyloglucanase transferases responsible for growth of plant cell wall, genes coding for the enzymes, a method of transferring xyloglucan molecules by using the enzyme, and methods of using the gene are described.
Claims6
An isolated and purified endo-xyloglucan transferase, which is encoded by a DNA comprising the DNA sequence shown in SEQ ID NO. 15.
An isolated and purified endo-xyloglucan transferase, which is encoded by a DNA comprising a DNA sequence whose complementary DNA sequence hybridizes with the DNA sequence shown in SEQ ID NO. 15.
An isolated and purified endo-xyloglucan transferase, having the following physicochemical properties:
- (1) action:
- splits a D-glucosyl linkage in a xyloglucan molecule and links the resultant reducing end of a xyloglucan molecular segment to D-glucose of the non-reducing end of another xyloglucan molecule;
- (2) substrate specificity;
- acts on xyloglucans derived from Vigna, Tamarind and Tropaeolum equally;
- acts neither on carboxymethyl cellulose nor on .beta.-(1,3) or .beta.-(1, 4) glucans, which act as excellent substrates for .beta.-1,4-glucanase;
- (3) optimum pH:
- about 5.8;
- (4) optimum temperature:
- about 30.degree. C.;
- (5) molecular weight:
- about 33,000 determined by SDS-polyacrylamide gel electrophoresis.
