Abstract
This application relates to nucleic acids encoding decay accelerating factor (hereinafter abbreviated as DAF), as well as vectors and cells which comprise such nucleic acids. Additionally, nucleic acids which encode variants of DAF, such as insertion, deletion or substitution variants, are described. This application also relates to the preparation of DAF in recombinant cell culture. In particular, it is concerned with the large scale manufacture of DAF suitable for pharmaceutical or diagnostic use.

cDNA encoding a polypeptide comprising the amino acid sequence for mature DAF as shown in FIGS. 1a-d.
Isolated nucleic acid encoding a polypeptide comprising the amino acid sequence for mature DAF as shown in FIGS. 1a-d.
Isolated nucleic acid encoding a polypeptide which comprises the amino acid sequence of mature DAF shown in FIGS. 1a-d with about 1 to 10 contiguous residues of the amino acid sequence deleted.
Isolated nucleic acid encoding a polypeptide comprising amino acid residues 1 through to 329 of the mature DAF amino acid sequence shown in FIGS. 1a-d.
Isolated nucleic acid encoding a polypeptide which is an insertion variant selected from the group consisting of:
- (Thr.sub.329 LeuLeu Cys.sub.330) mature DAF;
- (Arg.sub.100 His Arg.sub.101) mature DAF;
- (Lys.sub.125 GlnLys.sub.126 GlnLys.sub.127) mature DAF;
- (Pro.sub.193 LeuLeu Ala.sub.194) mature DAF;
- (Pro.sub.247 AspAspGlu.sub.248) mature DAF:
- (Thr.sub.202 SerSerThr.sub.283) mature DAF;
- and (Gly.sub.316 ThrThrThr.sub.317) mature DAF.
An isolated nucleic acid encoding a polypeptide which comprises the amino acid sequence of mature DAF shown in FIGS. 1a-d, wherein an amino acid has been substituted by another amino acid.
An isolated nucleic acid which encodes a polypeptide comprising the following amino acid sequence:
- AspCysGlyLeuProProAspValProAsnAlaGlnProAlaLeuGluGlyArgThrSerPheProGlu.
Nucleic acid encoding a polypeptide comprising the amino acid sequence:
- AspCysGlyLeuProProAspValProAsnAlaGlnProAlaLeuGluGlyArgThrSerPheProGlu, operably linked to a promoter which is heterologous to native DAF promoter.
Isolated nucleic acid encoding a polypeptide comprising the amino acid sequence for mature sDAF as shown in FIGS. 2a-e.
