## Abstract

A magnetically controllable, or guided, carrier composition and methods of use and production are disclosed, the composition for carrying biologically active substances to a treatment zone in a body under control of a magnetic field. The composition comprises composite, volume-compounded ferrocarbon particles of 0.2 to 5.0 .mu.m in size, and preferably between 1.0 and 4.0 .mu.m, containing 1.0 to 95.0% by volume of carbon, and preferably between about 20 and 50%. The particles are produced by a joint deformation of a mechanical mixture of iron and carbon powders. The obtained particles are placed in a solution of a biologically active substance to absorb the substance onto the particles. The combination is administered in suspension.

## Claims

### Claim 1

A method for localized in vive treatment of disease comprising:
providing a magnetically responsive ferrocarbon carrier adapted to pass through a blood vessel to a disease site bearing therewith a biologically active substance selected for its efficacy in treating a disease;
inserting a delivery means into the blood vessel of a body being treated for the disease to within a short distance from the site to be treated;
injecting said carrier through said delivery means; and
establishing a magnetic field exterior to the body and adjacent to said site of sufficient field strength to guide a portion of said carrier through the blood vessel to a point at or near the site so that a therapeutic amount of the biologically active substance contacts the disease site.

### Claim 2 (depends on 1)

The method of claim 1 wherein the active substance is absorbed on the carrier.

### Claim 3 (depends on 1)

The method of claim 1 wherein the biologically active substance is released from the carrier at the site by a process of replacement of the substance with a material having a higher specific gravity produced by the body.

### Claim 4 (depends on 3)

The method of claim 3 wherein the material is blood, and replacement of the substance by blood is less than 30% by volume.

### Claim 5 (depends on 1, 2, 3)

The method of claim 1, 2 or 3 further comprising the step that the carrier is removed from the body by excretion and metabolism.

### Claim 6 (depends on 5)

The method of claim 5 wherein the carrier is fully removed within 30 days of its injection.

### Claim 7 (depends on 1, 2, 3, 5)

The method of claim 1, 2, 3 or 5 wherein the carrier includes particles containing iron and carbon, with the carbon being distributed substantially uniformly throughout the volume of the particles.

### Claim 8 (depends on 1)

The method of claim 1, wherein the magnetic field established is as small as 250 oersteds/cm.

### Claim 9

A method for localized in vivo treatment of disease comprising:
absorbing a biologically active substance onto a magnetically responsive carrier composition including particles less than about 5 .mu.m in crossection, each particle containing carbon and iron with carbon distributed substantially uniformly throughout the volume of the particle;
injecting the carrier with the absorbed biologically active substance thereon into a body having a disease site;
establishing a magnetic field exterior to the body and adjacent to the site, wherein the magnetic field is of sufficient field strength to guide and retain at the site a portion of the carrier; and
releasing a therapeutic amount of the active substance from the carrier so as to contact the site.

### Claim 10 (depends on 9)

The method of claim 9 wherein the field strength of the established magnetic field is as low as 250 oersteds/cm.

### Claim 11 (depends on 9, 10)

The method of claim 9 or 10 wherein the amount of absorbed active substance is in excess of 2.5% by mass of the carrier.

### Claim 12 (depends on 9, 10)

The method of claim 9 or 10 wherein the amount of absorbed active substance is in excess of 7.5% by weight of the carrier.

### Claim 13 (depends on 9)

The method of claim 9 wherein the carrier is guided to soft tissue adjacent a network of blood vessels at the treatment site, thus avoiding substantial emboliization of any of the network of vessels.
