## Abstract

A novel mammalian cytokine, IL-11, and processes for producing it are disclosed. IL-11 may be used in pharmaceutical preparations for stimulating and/or enhancing cells involved in the immune response and cells involved in the proper functioning of the hematopoietic system.

## Claims

### Claim 1

An isolated IL-11 having the amino acid sequence of SEQ ID NO: 2 and having one or more of the following characteristics:
(1) an apparent molecular weight under reducing conditions on SDS PAGE of approximately 20 kd;
(2) a calculated molecular weight of approximately 20 kd;
(3) biological activity in a T1165 assay;
(4) biological activity in a megakaryocyte colony forming assay in the presence of IL-3;
(5) biological activity on a B cell plaque forming assay.

### Claim 2

An isolated mammalian IL-11 comprising amino acids 22-199 of SEQ ID NO: 2.

### Claim 3 (depends on 2)

An IL-11 of claim 2 comprising the human sequence.

### Claim 4 (depends on 2)

An IL-11 of claim 2 produced by culturing a cell transformed with a DNA sequence encoding expression of said IL-11 polypeptide in operative association with an expression control sequence capable of directing the replication and expression of said IL-11, and recovering from the conditioned medium thereof said IL-11 protein.

### Claim 5 (depends on 2)

A process for producing a mammalian IL-11 comprising culturing a cell line transformed with a cDNA sequence encoding a mammalian IL-11 of claim 2 in operative association with an expression control sequence capable of directing the replication and expression of said IL-11.

### Claim 6 (depends on 2)

An isolated DNA coding for an IL-11 of claim 2.

### Claim 7 (depends on 6)

A cell transformed with a DNA of claim 6 in operative association with an expression control.

### Claim 8 (depends on 7)

A cell according to claim 7 comprising a mammalian or bacterial cell.

### Claim 9 (depends on 6)

A plasmid vector comprising a DNA of claim 6.

### Claim 10 (depends on 2)

A pharmaceutical composition comprising a mammalian IL-11 of claim 2 in a pharmaceutically effective vehicle.

### Claim 11 (depends on 2)

Homogeneous mammalian IL-11 of claim 2 having biological activity in the T1165 assay in the absence of IL-6.

### Claim 12 (depends on 2)

A method for increasing platelet formation comprising administering an effective amount of IL-11 of claim 2.

### Claim 13 (depends on 12)

The method of claim 12, wherein said IL-11 is administered intravenously.

### Claim 14 (depends on 2)

An injectable pharmaceutical composition for increasing platelet formation comprising an aqueous solution of IL-11 of claim 2 and a pharmaceutically acceptable carrier.

### Claim 15 (depends on 6)

A DNA which hybridizes under stringent conditions of 0.1 x SSC at 65.degree. C. to a DNA of claim 6.
