Abstract
A human gastric lipase protein for use in the treatment of lipase deficiency. A process is described for producing gastric lipase using recombinant DNA technology to produce a host organism (for example E. coli) capable of producing a methionine-gastric lipase or precursor of the gastric lipase which may be cleaved to yield the gastric lipase. The host organism is transformed with a vector including a gene coding for a methionine-gastric lipase or a precursor of gastric lipase. The precursor protein is for example, pregastric lipase protein, or a fusion protein comprising gastric lipase and a heterologous protein. A pharmaceutical composition in unit dosage or liquid form is described.
Claims22
A process for producing a methionine-gastric lipase protein from human gastric mucosal tissue comprising the steps:
- providing an expression vector comprising a DNA sequence encoding a methionine-gastric lipase from human gastric mucosal tissue operably linked to transcriptional and translational control signals;
- transforming a host cell with said expression vector;
- culturing the transformed host under conditions such that the methionine-gastric lipase is produced;
- and
- isolating the methionine-gastric lipase produced.
A process for the production of a gastric lipase protein from human gastric mucosal tissue comprising the steps:
- providing an expression vector comprising a DNA sequence encoding a gastric lipase precursor protein from human gastric mucosal tissue operably linked to transcriptional and translational control signals;
- transforming a host cell with said expression vector;
- culturing the transformed host under conditions such that the gastric lipase precursor protein is produced;
- cleaving said gastric lipase precursor protein to produce gastric lipase protein;
- and
- isolating said gastric lipase protein.
An isolated DNA encoding a protein comprising the amino acid sequence of a gastric lipase protein from human gastric mucosal tissue.
