Abstract
Human calcitonin receptors have been cloned, sequenced and expressed by recombinant means. The receptors and antibodies thereto may be used in screening systems to identify agonists and antagonists of human calcitonin receptors, thereby providing means for treating and preventing abnormal bone resorption, as well as in methods of diagnosis.
Claims8
A method for identifying a compound which alters human calcitonin receptor mediated metabolism in a cell, which comprises:
- incubating the compound with eukaryotic cells transformed or transfected with a DNA sequence which directs expression of (a) a human calcitonin receptor polypeptide having the amino acid sequence shown in SEQ ID NO:
- 2, (b) a human calicitonin receptor polypeptide having the amino acid sequence shown in SEQ ID NO:
- 25, (c) a naturally occurring allelic variant of (a) or (b), or (d) a polypeptide comprising a N-terminal ligand-binding domain, a first transmembrane domain, and a first intracellular domain of (a), (b), or (c), monitoring the metabolism of the cells, and determining the effect of said compound on receptor-mediated metabolism in the cells.
