Abstract
A subgenomic promoter of a positive strand RNA virus is disclosed which directs the amplified expression of a structural gene in plant tissue. The core region and an upstream activating domain of the subgenomic promoter are identified. This promoter can be utilized in a modified virus, or in an appropriate engineered recombinant DNA derivative which may be chromosomally integrated or maintained as an episome in transformed cells.
Claims16
A method of using an artificially constructed recombinant DNA molecule comprising the nucleotide sequence of a (+) strand plant RNA virus subgenomic core promoter selected from the group consisting of tobamoviruses, tobraviruses, bromoviruses, cucumoviruses and alfalfa mosaic virus, wherein said (+) strand RNA virus comprises a consensus sequence consisting of the group AUCUAUGUU and sequences having a single mismatch thereto, and further comprising an undeviating domain corresponding to the nucleotide sequence from a BstXI site to the BglII site of the cDNA corresponding to the RNA of FIG. 2, and wherein said activating domain is positioned 5' to said subgenomic core promoter sequence, to express a plant-expressible structural gene in a transformable host plant cell when an RNA-dependent RNA polymerase of a Sindbis-like plant virus is present in the transformed host plant cell, comprising the steps of:
- (a) introducing said recombinant DNA molecule into said transformable host plant cell;
- and
- (b) maintaining said cell or progeny of said cell under conditions when said cell or said progeny transcribe a (-) strand RNA from said recombinant DNA molecule containing said subgenomic promoter and said plant-expressible gene, wherein said (-) strand RNA molecule is transcribed by RNA polymerase under the control of a polII promoter;
- transcribe said plant-expressible gene from said (-) strand RNA by an RNA-dependent RNA polymerase of a Sindbis-like plant virus, which RNA-dependent RNA polymerase recognizes said subgenomic promoter to form a (+) strand subgenomic transcript comprising said plant-expressible structural gene;
- and translate said (+) strand subgenomic transcript into protein;
- whereby said plant-expressible structural gene is expressed.
- (a) providing a host plant cell comprising said recombinant DNA molecule;
- and
- (b) growing said cell under conditions whereby said plant-expressible structural gene is expressed.
