Abstract
DNA segments encoding the Erwinia herbicola enzymes geranylgeranyl pyrophosphate (GGPP) synthase, phytoene synthase, phytoene dehydrogenase-4H and lycopene cyclase, vectors containing those DNA segments, host cells containing the vectors and methods for producing those enzymes and beta-carotene by recombinant DNA technology in transformed host organisms are disclosed.
Claims15
A purified and isolated DNA segment comprising a nucleotide sequence that contains at least 1125 base pairs that define the structural gene for the Erwinia herbicola enzyme lycopene cyclase that is present in a restriction endonuclease fragment comprising the approximately 1142 base pair Sph I-Bam HI restriction fragment of plasmid pARC1509, the approximately 1210 base pair Nco I-Pst I fragment of plasmid pARC147 or the approximately 1148 base pair fragment of plasmid pARC1520, said lycopene cyclase converting lycopene into beta-carotene.
A purified and isolated DNA segment comprising a nucleotide sequence that contains at least 1125 base pairs that define a structural gene for the Erwinia herbicola enzyme lycopene cyclase that is present in a plasmid selected from the group consisting of pARC1509, pARC147, pARC1520, pARC1512 or a DNA variant that has at least 80 percent identity to said gene and hybridizes with said gene under moderately high stringency conditions comprising hybridization at a temperature of about 50.degree.-55.degree. C. in 6XSSC and a final wash at a temperature of 68.degree. C. in 1-3XSSC, said nucleotide sequence encoding an enzyme that converts lycopene into beta-carotene.
A recombinant DNA molecule comprising a vector operatively linked to an exogenous DNA segment that contains at least 1125 base pairs defining a structural gene for the Erwinia herbicola enzyme lycopene cyclase that is present in a plasmid selected from the group consisting of pARC1509, pARC147, pARC1520 and pARC1512 or a DNA variant that has at least 80 percent identity to said gene and hybridizes with said gene under moderately high stringency conditions comprising hybridization at a temperature of about 50.degree.-55.degree. C. in 6XSSC and a final wash at a temperature of 68.degree. C. in 1-3XSSC, said nucleotide sequence encoding an enzyme that converts lycopene into beta-carotene, and a promoter for driving the expression of said enzyme in host organism cells.
Plasmid pARC1510 having ATCC accession number 40851.
Plasmid pARC1520 having ATCC accession number 40852.
Plasmid pARC1509 having ATCC accession number 40850.
