Abstract
The present invention provides an improved method of cryopreservation of cell suspensions and compositions useful in same wherein only a non-penetrating extracellular cryopreservation polymer is required.
Claims20
A method for cryopreservation of a cell suspension comprising the steps of:
- (a) mixing said cell suspension with a storage solution containing at least one non-penetrating polymer to produce a storage mixture wherein cells in said suspension contain an intracellular solution having a spontaneous intracellular freezing temperature of T.sub.hom and said storage solution has a glass transition temperature of T.sub.g, and
- (b) maintaining said storage mixture at a temperature above T.sub.hom and not more than T.sub.g, thereby avoiding ice nucleation and maintaining the intracellular solution in a supercooled state.
A method for the cryopreservation of a cell suspension comprising the steps of:
- (a) mixing said cell suspension with a storage solution containing at least one non-penetrating polymer to produce a storage mixture wherein cells in said suspension contain an intracellular solution having a spontaneous intracellular freezing temperature of T.sub.hom and
- (b) maintaining said storage mixture at or below a temperature where dehydration of said cells is effectively ceased, said temperature being higher than T.sub.hom thereby avoiding ice nucleation and maintaining the intracellular solution in a supercooled state.
