Abstract
A sensitive and accurate tissue culture system and kit fop detecting subtle changes in immune function is provided. The system is based on the comparison of IL-2 production by T helper cells in response to recall antigens including influenza A virus, tatanus toxoid, alloantigens, mouse xenogeneic antigens and the like or combinations thereof. Different stages of immune dysfunction can be differentiated and organ graft rejection can be predicted by the method of the present invention.
Claims1
A method for predicting organ transplant rejection, said method comprising:
- obtaining a sample of peripheral blood leukocytes from an organ transplant recipient;
- exposing said peripheral blood leukocytes to allogeneic peripheral blood leukocytes expressing HLA alloantigens, said allogeneic peripheral blood leukocytes having been depleted of antigen-presenting cells;
- and
- measuring the production of IL-2 by the CD4.sup.+ T helper cells of said peripheral blood leukocytes in response to said allogeneic peripheral blood leukocytes;
- wherein:
- non-responsiveness by the CD4.sup.+ T helper cells of said peripheral blood leukocytes being predictive of non-rejection of organ transplant;
- and
- responsiveness by the CD4.sup.+ T helper cells of said peripheral blood leukocytes being predictive of acute or chronic organ transplant rejection .
