Abstract
Novel shuttle vectors which are useful as shuttle vectors among microorganisms belonging to Escherichia coli, Gluconobacter and Acetobacter comprising one or more marker genes, a replication origin functional in Escherichia coli, a replication origin functional in Gluconobacter oxydans and a Mob site.
Claims8
A shuttle vector comprising a replication origin functional in Escherichia coli, a replication origin functional in Gluconobacter oxydans selected from the group consisting of a replication origin of Gluconobacter oxydans, a replication origin of an endogenous plasmid of Gluconobacter oxydans and a replication origin of an endogenous phage of Gluconobacter oxydans and a Mob site, wherein the vector is PGE1.
A transformed strain of Escherichia coli wherein said strain contains a shuttle vector, which vector comprises a replication origin functional in Escherichia coli, a replication origin functional in Gluconobacter oxydans selected from the group consisting of a replication origin of Gluconobacter oxydans, a replication origin of an endogenous plasmid of Gluconobacter oxydans and a replication origin of an endogenous phage of Gluconobacter oxydans and a Mob site, wherein the vector is PGE 1.
A transconjugate of a microorganism of the genus Gluconobacter or Acetobacter wherein said transconjugate contains a shuttle vector, the vector comprising a replication origin functional in Escherichia coli, a replication origin functional in Gluconobacter oxydans selected from the group consisting of a replication origin of Gluconobacter oxydans, a replication origin of an endogenous plasmid of Gluconobacter oxydans and a replication origin of an endogenous phage of Gluconobacter oxydans and a Mob site, wherein the vector is PGE 1.
A process for producing a transconjugate of a microorganism of the genus Gluconobacter or Acetobacter comprising conjugally mating the microorganism with a transformed or transfected Escherichia coli containing a shuttle vector wherein the vector comprises a replication origin functional in Escherichia coli, a replication origin functional in Gluconobacter oxydans selected from the group consisting of a replication origin of Gluconobacter oxydans, a replication origin of an endogenous plasmid of Gluconobacter oxydans and a replication origin of an endogenous phage of Gluconobacter oxydans and a Mob site, wherein the vector is PGE 1.
A process for producing a polypeptide comprising forming a shuttle vector, wherein the vector comprises a replication origin functional in Escherichia coli, a replication origin functional in Gluconobacter oxydans selected from the group consisting of a replication origin of Gluconobacter oxydans, a replication origin of an endogenous plasmid of Gluconobacter oxydans and a replication origin of an endogenous phage of Gluconobacter oxydans, a Mob site and a DNA sequence coding for the polypeptide operatively linked with an expression control sequence wherein the vector is PGE 1, transforming an Escherichia coli with the vector, transconjugating a microorganism of the genus Gluconobacter or Acetobacter with said Escherichia coli, and culturing the transconjugate to allow expression of the polypeptide, and recovering said polypeptide and recovering said polypeptide.
