Abstract
Eucaryotic NAD cyclases able to cause production of cyclic adenosine diphosphate ribose from nicotinamide adenine dinucleotide.
Claims16
A composition enriched for the eucaryotic NAD cyclase of the genus Aplysia, wherein said NAD cyclase is enriched by at least 10 fold compared to the NAD cyclase present in a gonad of the genus Aplysia, said NAD cyclase having an apparent molecular weight of 24,000-34,000 daltons after electrophoresis in a sodium dodecyl sulphate polyacrylamide gel under a reducing condition.
A purified NAD cyclase which causes production of cADPR from NAD, wherein said NAD cyclase naturally occurs in the genus Aplysia, and said NAD cyclase has an apparent molecular weight of 24,000-34,000 daltons after electrophoresis in a sodium dodecyl sulphate polyacrylamide gel under a reducing condition.
An essentially purified eucaryotic NAD cyclase, consisting essentially of a protein purified from the water soluble fraction of a gonad of the genus Aplysia, wherein said protein has enzymatic activity which causes production of cADPR from NAD, and said protein has an apparent molecular weight of 24,000-34,000 daltons after electrophoresis in a sodium dodecyl sulphate polyacrylamide gel under a reducing condition.
An essentially purified eucaryotic NAD cyclase obtainable by a process comprising the steps of:
- (a) adding the water soluble fraction of a gonad of the genus Aplysia to a cation-exchange resin,
- (b) collecting a first elution containing said NAD cyclase from said resin,
- (c) adding said first elution to a gel filtration column, and
- (d) collecting a second elution containing said NAD cyclase from said column, wherein said Second elution consists essentially of said NAD cyclase.
