Abstract
The present invention provides novel substituted opioid analgesic compounds of Formula I: ##STR1## which are opioid agonists, and which are useful as analgesic agents for the treatment of pain, pharmaceutical compositions comprising a therapeutically-effective amount of a compound of Formula I in combination with a pharmaceutically-acceptable carrier, and a method for eliminating or ameliorating pain in an animal comprising administering a therapeutically-effective amount of a compound of Formula I to the animal.

A compound of the formula:
- ##STR35## or a pharmaceutically-acceptable salt thereof, wherein:
- R.sup.1 is --CN;
- R.sup.2 is [--OR.sup.4 wherein R.sup.4 is ##STR36## alkyl;
- R.sup.3 is ##STR37## alkyl or alkoxy;
- X is O;
- and
- Y is hydrogen.
A compound of the formula:
- ##STR38## or a pharmaceutically-acceptable salt thereof, wherein:
- R.sup.1 is hydrogen;
- R.sup.2 is --OH;
- R.sup.3 is hydrogen;
- X is O;
- and
- Y is hydrogen.
A compound of the formula:
- ##STR39## or a pharmaceutically-acceptable salt thereof, wherein:
- R.sup.1 is C.sub.3 -C.sub.6 alkenyl;
- R.sup.2 is --OH;
- R.sup.3 is hydrogen;
- X is O;
- and
- Y is hydrogen.
A compound of the formula:
- ##STR40## or a pharmaceutically-acceptable salt thereof, wherein:
- R.sup.1 is ##STR41## R is --H or --C.sub.1 -C.sub.5 alkyl;
- n is an integer of from 1 to 5;
- R.sup.2 is --OH;
- R.sup.3 is hydrogen;
- X is O;
- and
- Y is hydrogen.
A compound having the structure:

A compound of the formula:

- ##STR46## or a pharmaceutically-acceptable salt thereof, wherein n is an integer of from 1 to 5.
A compound of the formula:
- ##STR47## or a pharmaceutically-acceptable salt thereof, wherein:
- R.sup.1 is C.sub.1 -C.sub.6 alkyl;
- and
- R.sup.4 is C.sub.1 -C.sub.5 alkyl.
A compound of the formula:
- ##STR48## or a pharmaceutically-acceptable salt thereof, wherein R.sup.1 is C.sub.1 -C.sub.6 alkyl.
A compound of the formula:
- ##STR49## or a pharmaceutically-acceptable salt thereof, wherein R.sup.1 is --A--B, wherein A is alkylene having one or two carbon atoms and B is phenyl, ##STR50##
A compound of the formula:
- ##STR51## or a pharmaceutically-acceptable salt thereof, wherein R.sup.1 is --C.sub.1 -C.sub.6 alkyl and Y is --CO--OH or --CO--O--C.sub.1 -C.sub.5 alkyl.
