## Abstract

Biological samples are analyzed for benzodiazepines in a single isocratic analysis using a chromatographic column system containing an immobilized enzyme reactor which cleaves glucuronic acid-conjugated benzodiazepines, an anion exchange column, a hydrophobic cation exchange column and a reverse-phase analytical column. Preferred methods of performing the analysis further involve the use of a hydrophobic cation exchange precolumn prior to the anion exchange column. The system readily lends itself to automation, automatic periodic sampling and benzodiazepine identification and quantification. The system is particularly well adapted to the determination and identification of benzodiazepines in urine samples.

## Claims

### Claim 1

A method for analyzing a biological sample for the presence of benzodiazepines, said method comprising:
(a) passing said sample through the following media in the sequence indicated:
(i) a contact medium comprised of an immobilized glucuronosohydrolase to cleave glucuronic acid from any species conjugated with said glucuronic acid;
(ii) a first separation medium comprised of an anion exchange medium, under conditions permitting retention of hydrophobic anions, with substantially no retention of benzodiazepines;
(iii) a second separation medium comprised of a hydrophobic cation exchange medium under conditions permitting retention of basic compounds of stronger basicity than benzodiazepines, with substantially no retention of benzodiazepines; and
(iv) a third separation medium comprised of a reverse-phase medium, under conditions permitting the chromatographic separation of benzodiazepines; and
(b) detecting said benzodiazepines individually upon their emergence from said third separation medium.

### Claim 2 (depends on 1)

A method in accordance with claim 1 in which said sample is passed through said first, second and third separation media by a carrier liquid comprised of an aqueous solution of a member selected from the group consisting of methanol, ethanol and isopropanol, at a concentration of from about 10% to about 50% by weight and at an approximately neutral pH.

### Claim 3 (depends on 1)

A method in accordance with claim 1 in which said sample is passed through said first, second and third separation media by a carrier liquid comprised of an aqueous solution of a member selected from the group consisting of methanol, ethanol and isopropanol, at a concentration of from about 30% to about 50% by weight and at an approximately neutral pH.

### Claim 4 (depends on 1)

A method in accordance with claim 1 in which said sample is passed through said first, second and third separation media by a carrier liquid comprised of an aqueous solution of ethanol at a concentration of from about 30% to about 50% by weight and at an approximately neutral pH.

### Claim 5 (depends on 1)

A method in accordance with claim 1 further comprising passing said sample through a preliminary separation medium prior to passing said sample through said first separation medium, said preliminary separation medium comprised of a hydrophobic cation exchange medium of a hydrophobicity sufficiently stronger than said second separation medium to retain any benzodiazepines in said sample, said sample being passed through said preliminary separation medium under conditions permitting retention by said preliminary separation medium of said benzodiazepines to the exclusion of a preliminary group of interfering species comprised of inorganic salts, proteins, peptides and hydrophilic compounds in said sample, then eluting said retained sample components free of said preliminary group of interfering species and thereby eliminating said interfering species from said sample prior to passage of said sample through said first separation medium.

### Claim 6 (depends on 5)

A method in accordance with claim 5 in which said passage of said sample through said preliminary separation medium is performed subsequent to said passage of said sample through said contact medium.

### Claim 7 (depends on 5)

A method in accordance with claim 5 in which said passage of said sample through said preliminary separation medium is comprised of:
(1) applying said sample to said preliminary separation medium with an application buffer comprised of a low ionic strength aqueous buffer at a pH of from about 5 to about 7;
(2) purging said preliminary separation medium with an aqueous alkaline buffer to remove therefrom acidic drugs present in said sample;
(3) purging said preliminary separation medium with an aqueous acidic buffer to increase interactions between basic compounds and said preliminary separation medium, thereby enhancing the ease of separation of basic compounds present in said sample from each other according to their degree of basicity; and
(4) eluting benzodiazepines from said preliminary separation medium preferentially relative to interfering compounds which are more basic than benzodiazepines by use of a carrier liquid comprised of an aqueous solution of a member selected from the group consisting of methanol, ethanol and isopropanol, at a concentration of from about 10% to about 50% by weight and at an approximately neutral pH.

### Claim 8 (depends on 7)

A method in accordance with claim 7 in which:
said aqueous alkaline buffer of step (2) is comprised of a buffer at a pH of from about 10 to about 12, containing from about 1% to about 10% by weight of a member selected from the group consisting of methanol, ethanol and isopropanol;
said aqueous acidic buffer of step (3) is comprised of a buffer at a pH of from about 4 to about 5.5; and
said carrier liquid of step (4) is comprised of an aqueous solution of a member selected from the group consisting of methanol, ethanol and isopropanol at a concentration of from about 30% to about 50% by weight.

### Claim 9 (depends on 7)

A method in accordance with claim 7 in which said preliminary separation medium is a polymeric medium with sulfonic acid functional sites.
