## Abstract

This invention relates to polypeptide antigens of T.ovis suitable for use in vaccines to protect ruminants against infection by cestode parasites. The antigens are preferably obtained by expression of DNA coding therefor in a recombinant host cell. Aspects of the invention include DNA encoding the antigens, vectors containing the DNA and hosts which express the antigens. The invention also provides antibody and DNA probes for use in identifying protective antigens of cestode parasites other than T.ovis.

## Claims

### Claim 1

An antigenic T. ovis polypeptide essentially free from T. ovis components occurring in nature and running as a 47-52 kDa doublet on SDS-PAGE, which reduces the severity of T. ovis infection in a ruminant.

### Claim 2 (depends on 1)

A composition suitable for reducing the severity of T. ovis infection in a ruminant, said composition comprising an effective amount of an antigenic polypeptide as claimed in claim 1 in association with a pharmaceutically acceptable carrier or adjuvant.

### Claim 3

An antigenic T. ovis polypeptide, 45W, essentially free from T. ovis components occurring in nature and running as a 47-52 kD doublet on SDS-PAGE, which reduces the severity of T.ovis infection in a ruminant, said antigenic polypeptide consisting essentially of the amino acid sequence: ##STR1##

### Claim 4 (depends on 3)

An antigenic polypeptide as claimed in claim 3, consisting of the amino acid sequence: ##STR2##

### Claim 5 (depends on 3)

A polypeptide as claimed in claim 3, which expressed in the host cell as a fusion protein.

### Claim 6 (depends on 3)

A polypeptide as claimed in claim 3, which expressed as a fusion protein with the enzyme glutathione s-transferase.

### Claim 7 (depends on 3)

A composition suitable for reducing the severity of T. ovis infection in a ruminant, said composition comprising an effective amount of an antigenic polypeptide as claimed in claim 3 in association with a pharmaceutically acceptable carrier or adjuvant.

### Claim 8

An antigenic T. ovis polypeptide, 45W, essentially free from T. ovis components occurring in nature and running as a 47-52 kDa doublet on SDS-PAGE and reducing the severity of T. ovis infection in a ruminant, said antigenic polypeptide consisting essentially of the amino acid sequence: ##STR3## said antigenic polypeptide 45W being produced by a process comprising (i) culturing a host cell containing an expression vector which encodes and expresses said polypeptide 45W, and
(ii) recovering said antigenic polypeptide 45W free of other naturally occurring T. ovis polypeptides.

### Claim 9 (depends on 1)

A polypeptide as claimed in claim 1, consisting of the amino acid sequence: ##STR4##

### Claim 10 (depends on 1)

A polypeptide claimed in claim 1, consisting of the amino acid sequence: ##STR5##

### Claim 11 (depends on 1)

A peptide fragment of a polypeptide as claimed in claim 1, which generates a protective response to T. ovis infection having a molecular weight of about 26 kDa calculated by SDS-PAGE.

### Claim 12 (depends on 9)

A peptide fragment of a polypeptide as claimed in claim 9, which generates a protective response to T. ovis infection having a molecular weight of 26 kDa calculated by ADS-PAGE.
