Abstract
The present invention discloses cationic lipids useful for making lipid aggregates for delivery of macromolecules and other compounds into cells. They are especially useful for the DNA-dependent transformation of cells. Also disclosed are lipids useful both for the delivery of macromolecules and also useful as intermediates for making other such lipids.

A composition having the structure
- wherein
- R.sub.1 and R.sub.2 independently of another, are C.sub.1-23 alkyl or alkenyl, or ##STR12## alkyl or alkenyl, Z.sub.1 and Z.sub.2 independently of one another, are H or unbranched alkyl C.sub.1-6, q is 1 to 6,
- X is selected from any of X.sub.1 -X.sub.8, X.sub.15 and X.sub.16 where
- X.sub.1 is --(CH.sub.2).sub.n Br, (CH.sub.2).sub.n Cl, (CH.sub.2).sub.n F, (CH.sub.2).sub.n F or (CH.sub.2).sub.n I where n=0-6 or
- X.sub.2 is --(CH.sub.2).sub.n NH.sub.2 n=0-6 or
- X.sub.3 is --NH--(CH.sub.2).sub.m --NH.sub.2 m=2-6 or
- X.sub.4 is --NH--(CH.sub.2).sub.3 --NH--(CH.sub.2).sub.4 --NH.sub.2 or
- X.sub.5 is --NH--(CH.sub.2).sub.3 --NH--(CH.sub.2).sub.4 --NH(CH.sub.2).sub.3 --NH.sub.2 or
- X.sub.6 is ##STR13## X.sub.7 is ##STR14## X.sub.8 is ##STR15## X.sub.15 is --(CH.sub.2).sub.r --SH where r is 0-6 or X.sub.16 is --(CH.sub.2).sub.s --S--S--(CH.sub.2).sub.t --NH.sub.2 where s is 0-6 and t is 2-6.
