Abstract
The present invention is directed to recombinant hosts which contain and express the AluI Type-II restriction endonuclease gene. The present invention is also directed to vectors or DNA molecules which contain the gene, and to methods of producing the AluI enzyme. One source of the enzyme is Arthrobacter luteus, although other microorganisms may be used to isolate restriction endonuclease isoschizomers of the invention.
Claims22
A recombinant host cell comprising an Arthrobacter gene coding for a restriction endonuclease, said restriction endonuclease is capable of recognizing the palindromic sequence:
- 5' AG.dwnarw.CT 3'
- 3' TC.uparw.GA 5'
- and cleaving said sequence between the deoxyguanidine and deoxycytidine residues.
A vector comprising an Arthrobacter gene coding for a restriction endonuclease, said restriction endonuclease is capable of recognizing the palindromic sequence:
- 5' AG.dwnarw.CT 3'
- 3' TC.uparw.GA 5'
- and cleaving said sequence between the deoxyguanidine and deoxycytidine residues.
A method of producing a restriction endonuclease which recognizes the palindromic sequence:
- 5' AG.dwnarw.CT 3'
- 3' TC.uparw.GA 5'
- and cleaves said sequence between the deoxyguanidine and deoxycytidine residues, said method comprising:
- (a) culturing a recombinant host cell comprising an Arthrobacter gene coding for said restriction endonuclease;
- and
- (b) isolating said restriction endonuclease from said host cell.
