2-amino-1, 4-dihydropyridine derivatives with calcium agonist and alpha.sub. -antagonist activity

A compound of the formula ##STR132## or a pharmaceutically acceptable salt thereof or an isomer or N-oxide thereof wherein:

• R.sup.2 is alkyl of 1-4 carbon atoms, CN, CH.sub.2 OH, CH.sub.2 OCH.sub.2 CH.sub.2 NH.sub.2 or NH.sub.2 ;
• R.sup.3 is NO.sub.2, H, CN, CONH.sub.2 or
• R.sup.2 and R.sup.3 taken together are:
• ##STR133## Cy, when R.sup.5 is CHO, CO.sub.2 CH.sub.2 CHOHC.sub.6 H.sub.5, CO.sub.2 CH(OCH.sub.3)C.sub.6 H.sub.5, NO.sub.2, CONHC.sub.6 H.sub.5, or CO.sub.2 alkyl of 1-10 carbon atoms, is a cyclic group selected from the group:
• ##STR134## R.sup.1 and R.sup.9 independently are H, alkyl of 1-4 carbon atoms, haloalkyl of 1-4 carbon atoms, alkoxy of 1-10 carbon atoms, halogen, or NO.sub.2 ;
• Cy, when R.sup.5 is COR.sup.7, CO.sub.2 R.sup.7 is phenyl optionally substituted with one or two substituents independently selected from the group consisting of alkyl of 1-4 carbon atoms, haloalkyl of 1-4 carbon atoms, alkoxy of 1-4 carbon atoms, thioalkyl of 1-4 carbon atoms, alkylsulfinyl of 1-4 carbon atoms,
• alkylsulfinyl of 1-4 carbon atoms, halogen, or NO.sub.2,
• X is NR.sup.4, O, S, SO, SO.sub.2 or N.fwdarw.O;
• Y is CH.sub.2 ##STR135## Z is ##STR136## where R.sup.4 is H or an alkyl group of 1-4 carbon atoms;
• R.sup.7 is ##STR137## where A is a straight or branched alkenyl, alkenylenyl, or alkylnylenyl chain or
• R.sup.10 is ##STR138## or 4-pyrimidinyl, R.sup.8 is Ar, ##STR139## where Ar is phenyl optionally substituted with one or two substituents independently selected from the group consisting of alkyl of 1-4 carbon atoms, haloalkyl of 1-4 carbon atoms, alkoxy of 1-4 carbon atoms, halogen, or NO.sub.2 ;
• Q is (CH.sub.2 )q, (CH.sub.2).sub.n O, (CH.sub.2).sub.n NH or (CH.sub.2).sub.n S;
• n is independently 1 to 4;
• q is 0 to 2;
• and
• r is 1 to 10 with the proviso that when R.sup.2 is alkyl of 1-4 carbon atoms, CN, CH.sub.2 OH, CH.sub.2 OCH.sub.2 CH.sub.2 NH.sub.2 or NH.sub.2 ;
• R.sup.3 is NO.sub.2, H, CN, CONH.sub.2,
• then
• R.sup.8 cannot be:
