Abstract
A mutation defining a gene for the N.sup..alpha. -acetyltransferase of yeast has been identified and cloned. Mutations in this gene have been constructed and used to produce cells which are substantially incapable of catalyzing N.sup..alpha. -acetylation of protein. Such cells are valuable tools for determining the amino acid sequence of uncharacterized proteins. Such cells are also valuable tools for expressing a recombinant protein lacking an acetyl group at its .alpha.-amino group.
Claims8
A prokaryotic or eukaryotic cell which expresses an altered AAA1-encoded N.sup..alpha. -acetyltransferase.
A recombinant molecule containing an altered AAA1 gene, wherein said recombinant molecule is DNA or cDNA.
A method for producing a peptide lacking an N.sup..alpha. -acetylated amino terminus which comprises expressing said peptide in a yeast cell having an AAA1 gene, wherein said gene contains a mutation resulting in the substantial loss of AAA1 gene product activity, and renders said cell unable to catalyze said N.sup..alpha. -acetylation of said peptide.
