Abstract
Polyionic derivatives of cyclodextrin polymers and cyclodextrins immobilized on a solid surface are disclosed. Compositions and methods for separating a molecular species, including but not limited to a biologically active protein, from a mixture, for the storage of protein factors and for the therapeutic biodelivery of protein factors which employ the polyionic derivatives of cyclodextrin polymers and cyclodextrins immobilized on a solid surface are also disclosed.
Claims24
A polyionic derivative of an alpha-, beta-, or gamma-cyclodextrin polymer in which the ionic group is sulfate, phosphate, carboxylate, or nitrate.
A chromatographic adsorption medium comprising (1) a polyionic derivative of an alpha-, beta- or gamma-cyclodextrin or a polymer thereof in combination with (2) a metal ion capable fo beign monovalent or divalent and immobilized on a solid surface.
A composition comprising a protein adsorbed on a polyionic derivative of an alpha-, beta-, or gamma-cyclodextrin polymer.
A method for purifying a proteinf from a mixture, comprising the steps of:
- providing a solid phase comprising a polyionic derivatized cyclodextrin,
- contacting said solid phase with said mixture under conditons in which said protein binds to said polyionic derivatized cyclodextrin and at least some contaminating components of said mixture do not bind to said polyionic derivatized cyclodextrin,
- removing said contaminating components from contact with said solid phase while said protein remains bound to said solid pahse, and
- contacting said solid pahse with a solution operable to release said protein from said derivatized cyclodextrin into said solution.
