Abstract
Highly purified antihemophilic factor can be produced from a cryoprecipitate by a process comprising a PEG precipitation step, a viral inactivation step and a gel filtration step, all steps being carried out at room temperature.
Claims6
A process for the production of a concentrate of antihemophilic factor (AHF) from cryoprecipitate comprising the steps of:
- (a) dissolving said cryoprecipitate;
- (b) removing non-AHF proteins by precipitation with polyethylene glycol (PEG);
- (c) treating said AHF with a chemical for viral inactivation;
- then
- (d) passing said AHF through a gel filtration column containing a size exclusion resin to remove said chemical and to isolate said AHF to at least 35 units of Factor VIII activity per ml of pooled concentrate;
- each of the steps (a) through (d) being carried out at a temperature of 20-25.degree. C.
