Abstract
This invention relates to improved methods for transposon tagging as a route to plant gene isolation. The specific improvements comprise efficient means for preselecting plants which have undergone transposition. Through use of this preselection, it is possible to substantially reduce the number of plants which must be inspected to locate individuals having the transposon excised and inserted into a gene of interest. The result is a substantial savings in time, costs and efficiency associated with growing and screening large numbers of plants for tagging and isolation of a desired plant gene.
Claims17
A method for isolating a plant gene of interest conferring a characteristic phenotype, wherein said method comprises:
- (a) transforming a plant carrying the gene of interest to make transformed plants by introduction of a DNA construct containing an AC transposon inserted within a selectable marker effecting resistance to streptomycin wherein the transposon renders the marker inoperable;
- and (b) isolating those transformed plants having the transposon excised from the marker and inserted into the plant gene of interest.
The method of claim 11 wherein the dicot is a species of Nicotiana.
A DNA construct comprising a DNA segment containing a selectable marker for use in plant cells, said marker effecting resistance to stroptomycin wherein the marker is operably linked to a plant promoter and further comprising an AC transposon positioned within the segment, said transposon rendering the marker inoperable.
A host cell containing a DNA construct comprising a DNA segment comprising a selectable marker effecting resistance to streptomycin, said marker for use in plant cells wherein the marker is operably linked to a plant promoter and comprises an AC transposon positioned within the segment rendering the marker inoperable.
