Methodology involving novel coupling reagents for the carboxyl-terminal amino acid sequence analysis of peptides and proteins is described.

In a process for the sequential degradation of a protein or peptide, which including reacting the carboxy terminus of said protein or peptide with a coupling reagent, the improvement which comprises utilizing, as the coupling reagent, a phosphoryl(thio)amide having the Formula A, B, C or D.
