Abstract
2-amino-5-(substituted or unsubstituted phenylalkyl)-1,3,4-thiadiazoles, e.g., 2-amino-5-(3-trifluoromethylbenzyl)-1,3,4-thiadiazole, prepared, e.g., by ring closure, of corresponding 1-(substituted or unsubstituted phenylalkanoyl)-thiosemicarbazide in a strong acid medium. The compounds are useful as minor tranquilizers and sleep inducers.
Claims3
A pharmaceutical composition for use in treating insomnia or anxiety comprising a therapeutically effective amount of a compound of the formula ##STR9## where R.sub.1, R.sub.2, and R.sub.3 each independently represent hydrogen, fluoro, chloro, or trifluoromethyl, and
- Z is -- (CH.sub.2).sub.n --, ##STR10## where R.sub.4 is lower alkyl, and
- n is 1, 2, 3, or 4, and
- m is 0, 1, 2, or 3, and
- p is 1, 2, or 3,
- provided that (a) at least one of R.sub.1, R.sub.2, and R.sub.3 is trifluoromethyl, and (b) that when more than one of R.sub.1, R.sub.2, and R.sub.3 is trifluoromethyl, they are bonded to other than adjacent carbon atoms and (c) where two of the substituents are trifluoromethyl, the remaining substituent is hydrogen, in association with a pharmaceutically acceptable carrier therefor.
A method of treating insomnia, which comprises administering to a mammal in need of said treatment an effective amount of a compound of the formula ##STR11## where R.sub.1, R.sub.2, and R.sub.3 each independently represent hydrogen, fluoro, chloro, or trifluoromethyl, and
- Z is -- (CH.sub.2).sub.n --, ##STR12## where R.sub.4 is lower alkyl, and
- n is 1, 2, 3, or 4, and
- m is 0, 1, 2, or 3, and
- p is 1, 2, or 3,
- provided that (a) at least one of R.sub.1, R.sub.2, and R.sub.3 is trifluoromethyl, and (b) that when more than one of R.sub.1, R.sub.2, and R.sub.3 is trifluoromethyl, they are bonded to other than adjacent carbon atoms and (c) where two of the substituents are trifluoromethyl, the remaining substituent is hydrogen.
A method of treating anxiety, which comprises administering to a mammal in need of said treatment an effective amount of a compound of the formula ##STR13## where R.sub.1, R.sub.2, and R.sub.3 each independently represent hydrogen, fluoro, chloro, or trifluoromethyl, and
- Z is -- (CH.sub.2).sub.n --, ##STR14## where R.sub.4 is lower alkyl, and
- n is 1, 2, 3, or 4, and
- m is 0, 1, 2, or 3, and
- p is 1, 2, or 3,
- provided that (a) at least one of R.sub.1, R.sub.2, and R.sub.3 is trifluoromethyl, and (b) that when more than one of R.sub.1, R.sub.2, and R.sub.3 is trifluoromethyl, they are bonded to other than adjacent carbon atoms and (c) where two of the substituents are trifluoromethyl, the remaining substituent is hydrogen.
