Abstract
A process for the manufacture of benzodiazepine derivatives which comprises reacting 2-amino-5-chloro-benzophenone with a primary amine H.sub.2 N--R.sub.1 to give an imine which is reacted with an amino-ester ##STR1## to give a ketimine said ketimine being then converted to a benzodiazepine derivative.
Claims10
A process for the manufacture of benzodiazepines of formula (IV) ##STR11## which process comprises:
- i. condensing 2-amino-5-chloro-benzophenone (I) with a ##STR12## primary amine R.sub.1 --NH.sub.2 to give an imine (II) ##STR13## ii. reacting the imine (II) with an amino ester ##STR14## to give a ketimine (III), ##STR15## and iii. converting the ketimine (III) to the benzodiazepine (IV) by cyclization,
- and in which R.sub.1 represents an Alk--NH.sub.2 group, an ##STR16## group or an Alk--OR.sub.4 group in which Alk denotes a straight or branched alkylene group with 2 to 6 carbon atoms and R.sub.3 and R.sub.4 each represent a C.sub.1 -C.sub.4 alkyl group and R.sub.2 represents a C.sub.1 -C.sub.4 alkyl group.
In a process for the manufacture of benzodiazepines which comprises condensing 2-amino-5-chloro-benzophenone with an amine to produce an imine, reacting the imine with an amino ester to produce a ketamine and converting the ketamine to said benzodiazepine by cyclization, the improvement wherein the 2-amino-5-chloro-benzophenone is reacted with an amine having the formula R.sub.1 --NH.sub.2 wherein R.sub.1 is Alk--NH.sub.2, ##STR18## or Alk--OR.sub.4 and Alk is a straight or branched alkylene group having 2 to 6 carbon atoms and R.sub.3 and R.sub.4 are alkyl groups having 1 to 4 carbon atoms.
A process for making a ketimine of the formula ##STR19## which comprises reacting an imine of the formula ##STR20## wherein R.sub.1 is Alk--NH.sub.2, ##STR21## or Alk--OR.sub.4 and Alk is a straight or branched alkylene group with 2 to 6 carbon atoms and R.sub.3 and R.sub.4 are alkyl groups of 1 to 4 carbon atoms with an amino ester having the formula ##STR22## wherein R.sub.2 is an alkyl group having 1 to 4 carbon atoms.
