Abstract
The present disclosure provides methods and compositions that may be used to detect and the presence of Coccidioides in a sample. By designing a proteome-wide peptide library covering covers both species of Coccidioides, new epitopes were found having antibody reactivity. A multi-peptide assay was used to validate the diagnostic utility of the discovered antigens. Thus, the disclosed technology reveals new Coccidioides targets for the development of diagnostics, vaccines and therapeutics.
Claims20
A method of detecting in a sample the presence of an antibody that binds to a Coccidioides protein, the method comprising:
- providing a biological sample from a subject suspected to be infected with Coccidioides;
- contacting the biological sample with a peptide comprising an amino acid sequence having at least 85% sequence identity to an amino acid sequence selected from the group consisting of SEQ ID NO:
- 1-50;
- and
- detecting antigen-antibody complexes formed.
An immunogenic composition, comprising:
- one or more peptides comprising an amino acid sequence having at least 85% sequence identity to an amino acid sequence selected from the group consisting of SEQ ID NO:
- 1-50;
- and
- a pharmaceutically acceptable carrier or adjuvant.
A kit for detecting the presence of Coccidioides in a subject comprising one or more peptides comprising an amino acid sequence having at least 85% sequence identity to an amino acid sequence selected from the group consisting of SEQ ID NOS:
- 1-50;
- and optionally, one or more of the following:
- (i) the peptides comprise an amino acid sequence having at least 85% sequence identity to an amino acid sequence selected from the group consisting of SEQ ID NO:
- 2-4, 8-9, 11-18, 23-25, 27, 29, 30-33, 35, 37-41, 43-44, and 48;
- (ii) the kit comprises a first, second, third, fourth, fifth, and/or sixth peptide, wherein the first peptide comprises an amino acid sequence having at least 85% sequence identity to an amino acid sequence selected from the group consisting of SEQ ID NO:
- 38, 39, 43, and 44, wherein the second peptide comprises an amino acid sequence having at least 85% sequence identity to an amino acid sequence selected from the group consisting of SEQ ID NO:
- 8-9, 11-18, 23-25, 27, and 29-33, wherein the third peptide comprises an amino acid sequence having at least 85% sequence identity to an amino acid sequence selected from the group consisting of SEQ ID NO:
- 2-4 and 35, wherein the fourth peptide comprises an amino acid sequence having at least 85% sequence identity to an amino acid sequence selected from the group consisting of SEQ ID NO:
- 40-41, wherein the fifth peptide comprises an amino acid sequence having at least 85% sequence identity to an amino acid sequence consisting of SEQ ID NO:
- 48, and/or wherein the sixth peptide comprises an amino acid sequence having at least 85% sequence identity to an amino acid sequence consisting of SEQ ID NO:
- 37;
- (iii) wherein the one or more peptides comprise an amino acid sequence having at least 85% sequence identity to an amino acid sequence selected from the group consisting of SEQ ID NO:
- 1, 3, 7, 13, 15, 17, 24, 26, 29, 32, 36, 38, 39, 40, 43, and 44;
- (iv) wherein the kit comprises 16 peptides comprising an amino acid sequence having at least 85% sequence identity to an amino acid sequence as set forth by SEQ ID NO:
- 1, 3, 7, 13, 15, 17, 24, 26, 29, 32, 36, 38, 39, 40, 43, and 44;
- (v) where each peptide is biotinylated;
- and/or (vi) wherein the kit comprises a plate having microwells coated with the peptides.
