Disclosed are systems, methods, and kits for enzyme-free, compartment-free digital detection of target analytes, such as single protein biomarkers and multivalent targets, using transient plasmonic nanobubbles (PNBs) generated around gold nanoparticles. In a bead-based embodiment, capture antibodies on microbeads bind the analyte, followed by labeling with detection antibody-conjugated gold nanoparticles. After washing, labeled complexes are flowed through a detection zone and exposed to pulsed laser light, generating individual PNBs detected as binary scattering signals via a probe laser and photodetector. Poisson statistics enable absolute quantification at attomolar sensitivity. In a homogeneous embodiment, pairs of antibody-conjugated nanoparticles aggregate upon binding multivalent targets, producing distinguishable PNB signals for rapid, wash-free detection. The platform offers high throughput, minimal sample volume (<10 μL), room-temperature operation, and simplified instrumentation for ultrasensitive biomarker and viral diagnostics.
