Abstract
Compositions and methods are provided for predicting the strength of an immune response that is elicited in humans following administration of a specific rAAV vector. The methods utilize antibody-opsonization of rAAV to enhance assays to reveal and quantify immune responses in human PBMC based assays.
Claims23
A method for determining the immunostimulatory potential of an AAV composition, the method comprising:
- combining in an assay (i) a test AAV composition;
- (ii) opsonizing anti-AAV antibodies;
- and (iii) a population of immune cells;
- incubating for a period of time sufficient to activate immune cells to release immune effector molecules;
- measuring the level of released immune effector molecules;
- wherein the level of immune effector molecule produced by the immune cells is indicative of the level of the immunostimulatory potential of the test AAV composition.
