Abstract
The invention provides methods and systems for designing and selecting primer pairs for targeted nucleic acid molecule amplification. The methods and systems of the invention enable detection and amplification of error-free molecules or molecules with specified genetic variations.
Claims11
A method of primer selection, the method comprising:
- selecting primer pairs from a database;
- making copies, using the selected primer pairs, from among a heterogeneous pool of DNA molecules;
- sequencing the copies to produce sequence data;
- mapping the sequence data to a target reference sequence, thereby generating mapping scores;
- and
- identifying, based on the mapping scores, a primer pair for amplifying the target reference sequence.
