In a method for detecting a carbapenemase-producing organism (CPO), a bacterial sample is lysed and the lysate is incubated with a carbapenem. The carbapenem is then transferred a solution including bacterial growth medium, an indicator bacterial strain that is susceptible to the carbapenem, a protease, and a cell viability indicator that can be metabolized by the indicator bacterial strain. After an incubation period, a change in the cell viability indicator confirms that the bacterial sample comprises a CPO, and no change in the cell viability indicator confirms that the bacterial sample does not include a CPO. A kit for an assay for detecting a CPO includes a lysis solution, a carbapenem, a bacterial growth medium, an indicator bacterial strain that is susceptible to the carbapenem, a protease, and a cell viability indicator that is metabolizable by the indicator bacterial strain.
