Abstract
Compositions of assembly solutions and stabilization solutions that can achieve kinetic control of the size and composition of polycationic/nucleic acid particles and the optimized process for reproducible and scalable production of concentrated, shelf-stable nanoparticle vehicles for in vitro transfection of cell culture in bioreactors are disclosed.
Claims35
A method for preparing a plurality of stable polycationic/nucleic acid complex nanoparticles, the method comprising:
- (a) providing one or more nucleic acids in a first buffer in a total nucleic acid concentration ranging from 0.1 to 0.8 mg/mL;
- (b) providing a polycationic polymer in a second buffer, wherein the first buffer and the second buffer can be the same or different;
- (c) mixing the polycationic polymer with the nucleic acid and incubating mixture for a period of time to form a plurality of polycationic/nucleic acid complexes;
- and
- (d) mixing a third buffer with the plurality of polycationic/nucleic complexes to form a plurality of stable polycationic/nucleic acid nanoparticles having a defined particle size.
