Abstract
A system, kit, and method for continuous directed evolution are provided, which include the use of a first expression construct comprising a SP6 promoter operably linked to a polynucleotide encoding a first protein of interest; and a second expression construct comprising at least one polynucleotide encoding a SP6 RNA polymerase-deaminase fusion protein.
Claims14
A system for continuous directed evolution, comprising
- (a) a first expression construct comprising a SP6 promoter operably linked to a polynucleotide encoding a first protein of interest;
- and
- (a) a second expression construct comprising at least one polynucleotide encoding a SP6 RNA polymerase-deaminase fusion protein.
A kit comprising
- (a) a polynucleotide encoding a SP6 RNA polymerase,
- (b) a polynucleotide encoding a cytosine deaminase and/or, a polynucleotide encoding an adenine deaminase, and
- (c) a SP6 promoter.
A method of continuous directed evolution of a protein of interest, comprising
- (a) introducing into a host cell a first expression construct comprising a SP6 promoter operably linked to a polynucleotide encoding the protein of interest;
- (b) introducing into the host cell a second expression construct comprising at least one polynucleotide encoding a SP6 RNA polymerase-deaminase fusion protein;
- and
- (c) culturing the host cell under conditions suitable for continuous directed evolution of the protein of interest.
