Abstract
The invention is drawn to an innovative approach that leverages stereolithography-based micropatterning to generate adhesive micro-colonies of human embryonic stem cells (hESCs) with defined size and geometry. These glass-supported micropatterned arrays provide precise spatial confinement, allowing for controlled self-organization of hESC-derived TE tissues.
Claims26
(canceled)
A method for producing trachea or esophagus organoids, the method comprising:
- seeding definitive endoderm cells onto rectangular microfabricated culture substrates comprising elongated confinement geometries, including rectangular and elliptical shapes, that spatially confines the cells;
- contacting the cells with a TGF inhibitor and a BMP inhibitor to obtain patterned foregut tissues;
- contacting the patterned foregut tissues with a culture medium cocktail comprising:
- KGF, BMP4, retinoic acid and a WNT agonist for tracheal tissue determination or
- KGF, Noggin, retinoic acid and a WNT agonist for esophageal tissue determination;
- and
- culturing the definitive endoderm, foregut and patterned foregut cells in complete serum-free medium to obtain tracheal or esophageal organoids.
4 more elements
A cultured tracheal or esophageal organoid derived from stem cells wherein:
- the tracheal organoids exhibit radial tissue anisotropy surrounding bud primordia, including an edge-enriched orientation bias (increased Edge-to-Center Ratio);
- and
- the esophageal organoids lack this feature.
