A method of preparing modified T cells, comprising modifying T cells during T cell activation by contacting the T cells with an inhibitory agent for prolyl hydroxylase, wherein the modifying optionally comprises increasing the expansion rate of T cells compared to unmodified T cells.
A method of producing Chimeric Antigen Receptor (CAR) T cells or T Cell Receptor (TCR)-T cells, comprising:
- (a) modifying T cells during T cell activation by contacting the T cells with an inhibitory agent for prolyl hydroxylase;
- (b) introducing a CAR transgene or a TCR transgene into the modified T cells of (a) to produce CAR-T cells or TCR-T cells;
- and
- (c) harvesting the CAR-T cells or TCR-T cells after (b).
