Abstract
The present disclosure is directed to methods of producing chimeric organoids containing cells of chimeric origin. The present disclosure is also directed to compositions comprising such chimeric organoids and the use of such chimeric organoids in high throughput screens.
Claims28
A method for making a chimeric organoid containing cells of chimeric origin, comprising:
- obtaining at least one embryoid body from each of at least two chimeric origins;
- producing a patterned organoid from each of the embryoid bodies;
- dissociating at least a portion of the cells from at least two of the patterned organoids to produce dissociated cells from each of the patterned organoids;
- mixing at least a portion of the dissociated cells produced in step (c);
- and
- culturing and expanding the mixed cells of step (d) under conditions and for a length of time sufficient to form a fully-formed brain organoid containing cells of chimeric origin.

A method of biobanking comprising:
- generating a population of dorsal forebrain organoids;
- dissociating the generated dorsal forebrain organoids at d15-d18 or d23-d25;
- and
- freezing the dissociated dorsal forebrain organoids comprising:
- transferring the dissociated dorsal forebrain organoids to a warmed solution containing papain enzyme,
- incubating the solution containing the dissociated dorsal forebrain organoids;
- mincing the solution to produce a single-cell suspension;
- mixing the single-cell suspension with an inhibitor solution;
- centrifuging the mixture and resuspending the resulting cell pellet in freezing medium;
- and
- freezing the resuspended mixture.
