The present invention provides a system for culturing cells. The system comprises a cassette, an insert having a cell culture platform, and cultured cells attached to or in the cell culture platform. Where the insert is in the cassette, the cultured cells may be exposed to a flowing fluid with a wall shear stress at, for example, 0.25-75 dyne/cm2. The cultured cells may be seeded and grown into, for example, a monolayer attached to the cell culture platform in the insert outside the cassette before the insert is placed in the cassette and the cultured cells are exposed to a flowing fluid. The insert may be removed from the cassette while the cultured cells remain attached to the cell culture platform in the insert. Also provided are a method for culturing cells in the system and a method for preparing the system.
