Abstract
An organ-on-chip system having a gradient of a stimulating agent (e.g., chemokine) and a plurality of immune cells expressing different phenotypes relative to their placement within the gradient and methods of using the same.
Claims32
An organ-on-chip system, comprising:
- a fluidic substrate having a plurality of channels and a chamber, the chamber being configured to house a hydrogel embedded with a plurality of immune cells or placed thereon;
- wherein the plurality of channels include at least one channel connected to the chamber to deliver a stimulating agent to the plurality of immune cells of the hydrogel;
- wherein the hydrogel comprises a gradient of the stimulating agent;
- and
- wherein different regions of the hydrogel each with different portions of the plurality of immune cells express different phenotypes relative to their placement within the gradient.
