Abstract
Provided are methods and compositions the identification of binding sites on nucleic acids in situ by particular proteins or protein complexes. The methods and compositions allow binding site mapping without the need for specific labeling, tagging, or engineering of the proteins or cells being assessed.
Claims31
An engineered, non-naturally occurring recognition-editing system comprising a fusion polypeptide comprising:
- (a) a protein-specific recognition module, wherein the module specifically recognizes a target protein;
- and
- (b) a nucleic acid base editor domain,
- wherein the nucleic acid base editor domain is capable of (i) altering one or more specific bases of a nucleic acid to a different base;
- or (ii) modifying a specific nucleoside of a nucleic acid in a manner that is detectable by nucleic acid sequencing.
