A β-elemene derivative containing a photoaffinity group diazirine, and a preparation method and use thereof as a photoaffinity molecular probe are provided. A compound having a structure shown in Formula (I), a pharmaceutical composition containing the compound having the structure shown in Formula (I), and a hydrate thereof, as well as an isotopic derivative, a chiral isomer, an allomer, different salts, prodrugs, and preparations of these compounds are provided. The preparation method and the use of the β-elemene derivative containing the photoaffinity group diazirine are also provided, where the compound has an anti-proliferation activity on various tumor cell lines, and is applied to screening of a β-elemene interacting protein. The β-elemene photoaffinity molecular probe is expected to become a molecular tool for targeted screening and action mechanism research of an elemene.
