The disclosure provides transmucosal film for delivering N,N-dimethyltryptamine (DMT) comprising an active layer comprising DMT or a pharmaceutically acceptable salt or prodrug thereof in a polymeric carrier matrix and a backing layer.

A transmucosal composition comprising:
- (a) an active layer comprising N,N-dimethyltryptamine (DMT) or a pharmaceutically acceptable salt or prodrug thereof in a polymeric carrier matrix, and
- (b) a backing layer,
- wherein the active layer comprises about 0.5 wt % to about 20 wt % of DMT or a pharmaceutically acceptable salt or prodrug thereof, and
- wherein the active layer comprises a plasticizer.
