A controller for a surgical microscope can control an excitation unit to emit excitation light for exciting fluorophores located in a target area of a patient, control an optical detection unit to receive light from the target area and separate the light into a first and a second spectral channels, generate an image of the target area, determine a first intensity for each pixel of the image based on the first spectral channel and a second intensity for each pixel based on the second spectral channel, and determine a first image region and a second image region based on the first intensities and the second intensities. The first image region corresponds to a first region of the target area exhibiting fluorescence caused by the excited fluorophores. The second image region corresponds to a second region exhibiting fluorescence caused by a fluorescence source other than the excited fluorophores.
