Abstract
A method for processing a sample of a cell culture composition includes diluting the sample followed by filtering the diluted sample, wherein the dilution may be at least 10 parts by volume of a diluent to one part sample, and the filter may be a porous membrane with a molecular weight cutoff of less than 20,000 Daltons. The dilution may be carried out at a multiple-port dilution valve having a first condition and a second condition. The sample may be mixed with the diluent at a predetermined dilution ratio when the dilution valve is in the second condition.
Claims12
A method for processing a liquid sample, the method comprising:
- (a) providing a processing apparatus having a dilution module including:
- (i) a first flow path;
- (ii) a second flow path;
- and
- (iii) a sample channel, wherein the dilution module is operable in first and second conditions, wherein the first and second flow paths intersect in both of the first and second conditions, and one or more of the first and second flow paths intersect with the sample channel only in the second condition;
- (b) conveying a first diluent flow of a diluent through the first flow path at a first flow rate;
- (c) conveying a second diluent flow of the diluent through the second flow path at a second flow rate that is different from the first flow rate;
- (d) conveying the liquid sample into the sample channel;
- and
- (e) adjusting the dilution module from the first condition to the second condition for a dilution time period when the sample is present in the sample channel to form a diluted sample.
A method for processing a sample having a liquid component, the method comprising:
- (a) providing a processing apparatus having:
- (i) a sample collection module including a first valve and a sample channel;
- (ii) a dilution module including a second valve operable in at least two conditions, with a first condition fluidically coupling a first port of the second valve to the first valve, and a second condition of the second valve fluidically coupling a second port of the second valve to the first valve while fluidically disconnecting the first port from the first valve;
- (b) conveying the sample into the sample channel;
- (c) conveying diluent through the second valve when the second valve is in the first condition, such that the diluent is flowable through the first valve into the sample channel through a connection channel between the first and second valves;
- (d) switching the second valve to the second condition, such that sample diluted with the diluent at the second valve is flowable to the first valve.
