Abstract
Disclosed are methods and compositions for detecting glomerular disease in dogs and effectively treating dogs with glomerular diseases such as amyloidosis, glomerulosclerosis, or immune complex-mediated glomerulonephritis. miRNAs associated with glomerular disease are also provided, as are kits, and methods of use.
Claims21
A method of detecting glomerular disease in a dog comprising:
- (a) obtaining a sample from the dog that comprises at least a first miRNA associated with glomerular disease;
- (b) detecting in the sample the presence of an elevated level of the miRNA relative to a control dog that lacks glomerular disease;
- wherein the elevated level of the miRNA relative to the control is indicative of glomerular disease in the dog.
A method of treating a dog with glomerular disease comprising:
- (a) obtaining a sample from the dog that comprises at least a first miRNA associated with glomerular disease;
- (b) detecting in the sample the presence of an elevated level of the miRNA relative to a control dog that lacks glomerular disease;
- and
- (c) treating the dog with glomerular disease based on the elevated level of the miRNA relative to the control.
A method of detecting the presence of a miRNA corresponding to glomerular disease in a sample, the method comprising:
- (a) contacting the sample comprising miRNA with a reverse transcriptase, wherein said reverse transcriptase produces a cDNA sequence corresponding to the miRNA;
- and
- (b) performing a polynucleic acid amplification reaction, thereby producing an amplicon;
- and
- (c) detecting said amplicon, wherein said amplicon comprises a DNA sequence produced from SEQ ID NO:
- 1-4 or 7.
The method of claim 17, wherein the method further comprises quantifying the amplicon.
A kit comprising:
- (a) a miRNA primer pair;
- (b) a means to quantitatively measure at least one miRNA;
- and
- (c) a container for the substrate.
