Abstract
The present invention relates to methods for producing Saccharomyces strains that are capable of growth on xylose as a sole carbon source at a desired growth rate, (such as at least one generation per 48 hours), strains made by such methods, and Saccharomyces strains that grow at a growth rate of at least one generation per 48 hours using xylose as a sole carbon source for growth made by non-recombinant methods.
Claims32
A genetically engineered yeast cell, the engineered yeast cell comprising:
- a deletion or disruption of a native gene encoding an erythrose reductase enzyme at least 60%, at least 65%, at least 70%, at least 75%, at least 80%, at least 85%, at least 90%, at least 95%, at least 98%, at least 99%, or 100% identical to at least one of SEQ ID NOs:
- 63 and 66.
The yeast cell of any preceding claim, wherein the yeast cell is selected from the group consisting of Trichosporonoides megachiliensis, Trychosporonoides oedocephalis, Trychosporonoides nigrescens, Pseudozyma tsukubaensis, Trigonopsis variabilis, Moniliella, Ustilaginomycetes, Trichosporon, Yarrowia lipolytica, Saccharomyces cerevisiae, Penicillium, Torula, Pichia, Candida, Candida magnoliae, and Aureobasidium.
The yeast cell of any preceding claim, wherein the yeast cell is a Moniliella pollinis cell and the gene encoding the erythrose reductase enzyme is at least 60%, at least 65%, at least 70%, at least 75%, at least 80%, at least 85%, at least 90%, at least 95%, at least 98%, at least 99%, or 100% identical to at least one of SEQ ID NOs:
- 61 and 64.
The yeast cell of any preceding claim, wherein, when the engineered cell is used in a fermentation process in the presence of dextrose, titer and/or yield of erythritol production is decreased relative to titer and/or yield of erythritol in an equivalent fermentation process using an equivalent cell in which there has been no disruption or deletion of a native gene encoding an erythrose reductase at least 60%, at least 65%, at least 70%, at least 75%, at least 80%, at least 85%, at least 90%, at least 95%, at least 98%, at least 99%, or 100% identical to at least one of SEQ ID NOs:
- 63 and 66.
The yeast cell of any preceding claim, wherein at least one copy of the gene encoding the erythrose reductase enzyme has been replaced with a native or exogenous polynucleotide sequence.
A genetically engineered Moniliella pollinis cell comprising a deletion or disruption a native gene encoding an erythrose reductase enzyme at least 60%, at least 65%, at least 70%, at least 75%, at least 80%, at least 85%, at least 90%, at least 95%, at least 98%, at least 99%, or 100% identical to at least one of SEQ ID NOs:
- 63 and 66.
A method of fermentation comprising
- contacting a substrate comprising dextrose with the engineered yeast cell of any preceding claim, wherein fermentation of the substrate by the engineered cell produces less erythritol than fermentation using an equivalent yeast cell that does not have a deletion or disruption of said ER encoding gene.
