A trilaminar membrane invention to quantitate lactose in conjunction with a potentiostatic amperometric cell. The invention features a support layer, an immobilized enzyme layer, and an electrode-facing exclusion layer. The support layer is a homogeneous, semi-permeable polycarbonate film that controls diffusion rates of analyte into the enzyme reaction layer. The analyte is then oxidatively processed to generate hydrogen peroxide (H2O2) by the immobilized 2-enzyme system comprised of β-galactosidase and glucose oxidase. Hydrogen peroxide then diffuses through the permselective exclusion layer to react at a poised sense electrode probe to generate an electrical signal for analyte quantitation. The thin sensing membrane is affixed to an O-ring for mounting on the face of the electrode probe.
