Abstract
Disclosed are compositions and methods for targeted treatment of glioblastomas. In particular, bi-specific chimeric antigen receptor (CAR) T cells are disclosed that can be used with adoptive cell transfer to target and kill glioblastomas. Therefore, also disclosed are methods of providing an anti-tumor immunity in a subject with glioblastomas that involves adoptive transfer of the disclosed bi-specific CAR T cells.
Claims29
A method of providing an anti-cancer immunity in a subject with a cancer expressing IL13rα2 and EGFRviii, the method comprising administering to the subject an effective amount of the bi-specific chimeric antigen receptor (CAR) T cell of claim 25, thereby providing an anti-tumor immunity in the subject.
2-19. (canceled)
A bi-specific chimeric antigen receptor (CAR) T cell, comprising an immune effector cell engineered to express (a) a CD40 agonist and (b) a chimeric antigen receptor polypeptide that selectively binds IL13rα2 and EGFRviii.
An expression vector, comprising a nucleic acid sequence encoding a CD40L polypeptide and a nucleic acid sequence encoding a bi-specific chimeric antigen receptor (CAR) polypeptide that selectively binds IL13rα2 and EGFRviii, operably linked to an expression control sequence.
