Pharmaceutical compositions of colchicine for oral administration are provided for use in the treatment or prevention of atherosclerotic cardiovascular disease. The compositions comprise a sustained-release component, an immediate-release component, or a combination thereof, which are selectively adjustable and deliver colchicine according to a predetermined or desired release profile. The disclosed compositions and methods are designed to reduce atherosclerotic plaque, including reducing total plaque burden, and influencing plaque composition, such as non-calcified, fibrous, fibro-fatty, and calcified plaque, through targeted inhibition of vascular inflammation. In certain embodiments, administration of colchicine results in measurable changes in plaque volume and/or composition as assessed by imaging modalities, including coronary computed tomography angiography (CCTA) or intravascular ultrasound (IVUS). Methods of reducing plaque burden, altering plaque characteristics, and stabilizing vulnerable plaques, thereby lowering the risk of acute cardiovascular events in mammalian subjects, are also provided.
A method for reducing atherosclerotic plaque in a human subject, thereby reducing the risk of a cardiovascular event, which comprises:
A method for treating atherosclerotic cardiovascular disease in a human subject, which comprises administering to the subject a daily dose of about 0.25 mg to about 0.75 mg of colchicine, wherein said administration results in a reduction in total plaque volume as measured by coronary computed tomography angiography (CCTA) or intravascular ultrasound (IVUS).
A method of treating a subject with atherosclerotic cardiovascular disease, comprising:
A method for reducing atherosclerotic plaque in a human subject, the method comprising:
