A cooling chamber for a portable temperature-controlled enclosure is disclosed, the cooling chamber comprising a payload chamber defining an interior volume configured to receive temperature-sensitive contents. The payload chamber includes an aluminum structural wall having a hydrocarbon-based thermal control medium bonded thereto such that the aluminum structural wall functions as both a load-bearing element and an active thermal interface. A conductive grill structure comprising aluminum and having the hydrocarbon-based thermal control medium bonded thereto is positioned adjacent to the payload chamber and is configured to redistribute heat laterally across a surface area greater than that of the payload chamber wall. A heat extraction coupling interface is thermally coupled to the payload chamber and the conductive grill structure and is configured to conduct heat away from the payload chamber along a defined thermal pathway. A metallic heat extraction block having higher thermal conductivity than the aluminum structural wall is thermally coupled to the heat extraction coupling interface and is configured to passively spread and stabilize heat received from the payload chamber. A polymeric thermal isolation enclosure surrounds the payload chamber, conductive grill structure, and heat extraction coupling interface, the components cooperating to establish a layered and directional thermal pathway for regulating temperature within the interior volume.
