Abstract
Provided herein are compositions and methods of hyaluronan biosynthesis, secretion and length control.

A method to synthesize Hyaluronan (HA) and modulate HA's length, alter catalytic rate of HA production and/or alter polydispersity of HA comprising expressing a mutant nucleic acid sequence coding for mutant HA synthase (HAS) in a host cell, wherein the mutant HA synthase has one or more substitutions in WGTSGRK/R motif or WGTRG motif.
A method to synthesize Hyaluronan (HA) and modulate HA's length, alter catalytic rate of HA production and/or alter polydispersity of HA comprising expressing a mutant nucleic acid sequence coding for mutant HA synthase (HAS) in a host cell, wherein the mutant HA synthase has one or more substitutions in a transmembrane (TM) channel.
A method to synthesize Hyaluronan (HA) and modulate HA's length, alter catalytic rate of HA production and/or alter polydispersity of HA comprising expressing a mutant nucleic acid sequence coding for mutant HA synthase (HAS) in a host cell, wherein the mutant HA synthase has one or more substitutions in the active site channel, wherein the substitutions are at C307 or C337.
