Abstract
The present invention includes compositions and methods comprising CART cells with SHP-1 and/or SHP-2 genes knocked out.
Claims20
A genetically modified cell comprising a chimeric antigen receptor (CAR),
- wherein the CAR comprises an antigen binding domain, a transmembrane domain, and an intracellular domain, and
- wherein at least one gene selected from the group consisting SHP-1 and SHP-2, has been modified in the cell, wherein the SHP-1 and/or SHP-2 modification is carried out by a CRISPR-Cas9 system comprising at least one guide RNA (gRNA) that targets SHP-1 and/or SHP-2.
A method of treating cancer in a subject in need thereof, the method comprising administering to the subject a T cell genetically engineered to express a CAR,
- wherein the CAR comprises an antigen binding domain, a transmembrane domain, and an intracellular domain, and
- wherein at least one gene selected from the group consisting SHP-1 and SHP-2, has been modified in the T cell, wherein the SHP-1 and/or SHP-2 modification is carried out by a CRISPR-Cas9 system comprising at least one guide RNA (gRNA) that targets SHP-1 and/or SHP-2.
